L-Carnitine ((R)-carnitine) is a quaternary ammonium compound that serves as the essential carrier molecule of the mitochondrial carnitine shuttle, the transport system responsible for moving long-chain fatty acids into the mitochondrial matrix for β-oxidation. Through sequential interactions with carnitine palmitoyltransferase I (CPT1), carnitine-acylcarnitine translocase (CACT), and carnitine palmitoyltransferase II (CPT2), L-Carnitine enables the conversion and transport of long-chain acyl groups across the inner mitochondrial membrane. Because this shuttle represents a key regulatory step in fatty acid utilization, L-Carnitine is widely used in research investigating mitochondrial bioenergetics, fatty acid oxidation pathways, CPT1/CPT2 enzyme activity, acylcarnitine metabolism, metabolic flux analysis, and OCTN2 (SLC22A5) transporter function. It is also commonly utilized in cell-based and tissue-based studies examining mitochondrial substrate utilization and acylcarnitine profiling.
